In a sweet surprise to his fans in Japan in 2012, Siwan read out a letter in Japanese.
He starts off serious but brings in his signature wit towards the end. Overall it's a beautiful message indeed.

Update: I was intrigued why everyone laughed, it seemed like an inside joke. I couldn't understand the song as well.

Siwan said
At 2:45, "That's enough" ("ただもういい" ?). People began to laugh. I don't know why exactly. Perhaps because he sounded like he was going to say something bigger, but there was no more, just stopped there.
At 2:54, "だーかーらー" (Da ̄ ka ̄-ra ̄ ) (roughly, "the - re - fore -"), then
At 2:56, "ええじゃないかええじゃないか" (Ei Ja Nai Ka)
The phrase is dance, rhythm, song, and chant in celebration. So the other group members came forward to chant and dance together. The phrase's meaning is both defiant and fatalistic, and it translates as "Who cares?", "Why not?" or "What the hell?" (https://en.wikipedia.org/wiki/Ee_ja_nai_ka)
I think it's the sudden change of mood that makes it funny 😉
